dc.description.abstractBirçok antikanser ilaç ve kimyasalın hücre içi hipoksik bir ortam yaratarak kanser hücrelerinde hücre ölümüne neden olduğu bilinmektedir. Hücrelerin ekzojen oksidanlar veya serbest radikallerle inkübasyonunun hücre ölüm sürecini tetiklediği gösterilmiştir. Hücrede stres yanıtı diye bilinen yanıta neden olan bu etmenler, hücreden hücreye değişen konsantrasyonlarda, hücre içi sinyal ileti mekanizmalarının aktivasyonu şeklinde ilerleyen olayların başlatılması ile ya kontrollü hücre ölümü olarak bilinen apoptoza, veya nekroza yol açarlar. Bu çalışmada, hipoksinin kanser hücrelerine olan etkisini incelemek için insan meme kanser hücre soyu olan MDA-MB 23 1 hücrelerini, ve hücre içi hipoksi yaratmak için hidrojen peroksit (H2O2) kullandık. H2O2, konsantrasyon ve zamana bağlı olarak MDA-MB 231 hücrelerinde ölüme yol açtı. Hücre ölümünü karakterize etmek için MDA-MB 23 1 hücreleri DNA-spesifık bir boya olan D API ile boyanarak floresan mikroskopta incelendi. İmM H2O2 ile 24 saat inkübe edilmiş hücrelerde DAPI ile boyanan kromatinin yoğunlaşması ve nükleer fragmanları içeren apoptotik cisimcikler oluşması, ve sitoplazmanın büzülmesi gibi apoptotik hücre ölümünün karakteristik özellikleri görüldü. Apoptotik hücre ölümünde DNA hasarını gösteren DNA fragmantasyonu 24 saat H2O2 uygulanan hücrelerden elde edilen genomik DNA'nın agaroz jel elektroforezi ile analizinde belirgin olarak gözlendi. Bu bulgular, MDA-MB 231 hücrelerinin H202 ile yaratılan hipoksiye diğer tümör hücrelerine göre daha yüksek konsantrasyonlarda cevap verdiğini ve kemoterapi amacı ile yaratılan hipoksinin aynı zamanda bazı kanser hücrelerinde ilaçlara karşı direnç de yaratabileceğini düşündürmektedir.en_US
dc.description.abstractIt is well known that many anticancer drugs and chemicals can cause cell death via induction of intracellular hypoxic environment. It has been shown that incubation of the cells with exogenous oxidants or free radicals induces cell death process. These factors cause stress response in the cells, and in the concentrations varying from one cell to another, lead to either programmed cell death, which is also known as apoptosis, or necrosis, by activation of intracellular signal transduction mechanisms. In this study, we have used MDA-MB 231 human breast cancer cell line to investigate the effect of hypoxia on cancer cells, and to create intracellular hypoxia, hydrogen peroxide (H2O2 ) was used. H2O2 caused concentration- and time-dependent cell death in MDA-MB 23 1 cells. To further characterise the cell death, MDA-MB 23 1 cells were examined under fluorescent microscope after staining with a DNA-specific dye, DAPI. After 24 incubation with 1 mM H2O2, the characteristics of apoptotic cell death, such as condensation of D API- stained chromatin, appereance of apoptotic bodies which contain nuclear fragments, and shrinkage of cytoplasm were observed. In the analysis of genomic DNA from the cells treated with H2O2 for 24 h in agarose gel electrophoresis, DNA fragmentation, which is the indicator of DNA damage in apoptotic cells was observed. These findings indicate that, MDA-MB 231 cells respond to H2O2 -induced hypoxia at higher concentrations compared to other tumor cells, and imply that hypoxia created during chemotherapy may cause resistance to chemotherapeutic drugs.en_US
