dc.description.abstractSpermiyogram testi yaptırmak için merkezimize başvuran 50 hastanın semen parametreleri dondurma öncesi ve sonrası olmak üzere değerlendirildi. Normal spermatozoon oranı dondurma işlemi öncesi ve sonrası Papanicolau boyama yöntemi ile boyanarak saptandı. Dondurma sonrasında normal spermatozoon oranında % 60,4 azalma saptandı. Dondurma işlemi sonrası görülen bu azalma istatistiksel olarak da anlamlı bulundu. Total motil spermatozoon sayısı Makler kamarada dondurma işlemi öncesi ve sonrası değerlendirildi, dondurma sonrası motilite oranında % 46,2' lik azalma tespit edildi. Dondurma işlemi sonrası görülen bu azalma istatistiksel olarak da anlamlı görüldü. Gradient yöntemi ile yıkama sonrası spermatozoon tarafından üretilen bazal reaktif oksijen türlerinin düzeyleri, luminole ve lusigene bağımlı kemilüminesans yöntemleri ile araştırıldı. Dondurma işlemi sonrası örneklerde % 82.1 oranında artmış ROT üretimine rastlandı. Luminole ve lusigene bağımlı kemilüminesans değerleri dondurma işlemi sonrası örneklerde, taze örneklere oranla anlamlı derecede yüksekti. Çalışmamızda, taze ve dondurma işlemi sonrasında seminal plazma antioksidanlarından süperoksit dismutaz, katalaz, glutatyon peroksidaz ve total sülfidril gruplarının seviyeleri de araştırıldı. Seminal plazma antioksidan gruplarına ait değerlerin tümü, dondurma işlemi sonrası taze seminal plazma değerlerine göre anlamlı derecede düsmüs olarak bulundu. Dondurma işlemi sonucunda seminal plazma reaktif oksijen türlerinin düzeylerinde artma ve seminal plazma antioksidanlarının düzeylerinde azalma tespit edildi, bu değişikliklerin spermatozoon normal morfolojisi ve total motilitesi üzerine etkili olduğu saptandı. Bu bulgular ışığında, spermatozoonu etkileyen parametrelerdeki bozuklukların fertiliteyi de etkiliyebileceği sonucu çıkarıldı.en_US
dc.description.abstractThe fresh and cryopreserved semen samples of 50 patients applied to the ART (Asissted Reproductive Techniques) have been evaluated. The percentage of normal sperm were assiessed by dying with Papanicolau method before and after cryopreservation. After cryopreservation 60.4 % of decrease was detected in the percentage of normal spermatozoa. Also, the total number of motil spermatozoa was examined inboth fresh and cryopreserved samples and a 46.2 % of decrease was noted. The levels of basal reactive oxygene species in seminal plasma were investigated by chemiluminiscence related luminale and lusigene methods. In the samples obtained after cryoprocedure the ROS production was noted to be increasead by 82.1%. The values of chemiluminiscence related luminale and lusigene were meaningfully high in fresh samples compared to the cryopreserved samples. In addition, we have investigated the levels of seminal plasma SOD, catalase, glutation peroxidase and total sulphydride groups in fresh and cryopreserved samples. The levels of seminal plasma antioxidants were found meaningfully low after cryopreservation in comparison with the fresh samples. All together these findings revealed that cryopreservation damaged the morphology, total motility and the levels of basal reactive oxygen species and seminal plasma antioxidants, which may lead to infertility.en_US
