METHODS A qualitative open-ended survey was conducted in Istanbul and Ankara in 2021 with 58 respondents from 3 different districts in each city from four types of venues: restaurants, traditional coffee and waterpipe houses, and European-style cafes. The open-ended survey included questions to understand the knowledge, beliefs, and attitudes of respondents about Turkiye's smoke-free policy and their perceptions of the facilitators and/or barriers to smoke-free policy implementation and changes after COVID-19. The data were analyzed using an inductive approach to identify patterns and categorize the data into themes. RESULTS The respondents expressed that the smoke-free policy aimed to protect employees and customers from secondhand smoke (SHS), respect human health, and improve air quality. Findings suggest that the positive attitude of venue owners and staff toward the smoke-free policy serves as a facilitator. However, fear of financial impact, customers' negative attitudes, difficulties in meeting physical requirements, and insufficient enforcement were found to be barriers to implementing the smoke-free policy. The effects of the COVID-19 pandemic were reported as an initial increase in compliance and awareness among customers and staff, but some respondents noted negative changes due to the emotional and financial effects of prolonged restrictions. These challenges have led to decreased attention on the smoke-free policy among venue owners, staff and customers. Respondents' suggested improvements were related to building infrastructure, such as the ventilation systems and educating the public on the harmful health effects of smoking. CONCLUSIONS Despite the general understanding of the dangers of secondhand smoke and the smoke-free policy, this study highlights the challenges in implementing smoke-free policy measures and the continued need to raise awareness about the importance of a 100% smoke-free venue. Steady-state fluorescence (SSF) technique was employed for studying sol-gel transition and swelling of PAAm-kappa C composite gels which were prepared by free-radical crosslinking copolymerization. Pyranine was introduced as a fluorescence probe. Pyranine molecules start to bind to acrylamide polymer chains upon the initiation of the polymerization thus the spectra of the bonded pyranines shift to the shorter wavelengths. Fluorescence spectra from the bonded pyranines allow one to monitor the sol-gel transition and to test the universality of the sol-gel transition as a function of some kinetic parameters like polymer concentration. Observations around the gel point t (c) for PAAm-kappa C composite gels showed that the gel fraction exponent beta obeyed the percolation result for low kappa C (< 2.0 wt. %) however classical results were produced at higher kappa C (> 2.0 wt.%). On the other hand fluorescence intensity of pyranine was measured during in situ swelling process at various amounts of kappa C and it was observed that fluorescence intensity values decreased as swelling is proceeded. Disc shaped gels were formed from acrylamide (AAm) and N N'- methylenebisacrylamide(Bis) with various kappa- carrageenan (kappa C) contents by free radical crosslinking copolymerization in water. Pyranine (P) was doped as a fluorescence probe and scattered light I (sc) and fluorescence intensities I were monitored during drying of these gels. It is observed that fluorescence intensity of pyranine increased as drying time is increased for all samples. The increase in I was modeled using Stern- Volmer equation and diffusion with moving boundary. It is found that desorption coefficient D decreased as kappa C contents were increased. A docking protocol was established in order to classify each receptor conformation via its binding affinity to selected ligands with known efficacy. This work's main goal was to reveal many subtle features of the ligand-binding site presenting alternative conformations which might be considered as either active- or inactive-like but mostly specific for that ligand. Agonists inverse agonists and antagonists were docked to each MD conformer with distinct binding pockets using different docking tools and scoring functions. Mostly favored receptor conformation persistently observed in all docking/scoring evaluations was classified as active or inactive based on the type of ligand's biological effect. Here we studied the structure of a member of the Cys-loop ligand gated ion channel protein superfamily of receptors human type A gamma(2)alpha(1)beta(2)alpha(1)beta(2) gamma amino butyric acid receptor complex in a lipid bilayer environment. Studying the correlation between the structure and function of the gamma amino butyric acid receptor may enhance our understanding of the molecular basis of ion channel dysfunctions linked with epilepsy ataxia migraine schizophrenia and other neurodegenerative diseases. The structure of human gamma(2)alpha(1)beta(2)alpha(1)beta(2) has been modeled based on the X-ray structure of the Caenorhabditis elegans glutamate-gated chloride channel via homology modeling. The template provided the first inhibitory channel structure for the Cys-loop superfamily of ligand-gated ion channels. The only available template structure before this glutamate-gated chloride channel was a cation selective channel which had very low sequence identity with gamma aminobutyric acid receptor. Here our aim was to study the effect of structural corrections originating from modeling on a more reliable template structure. The homology model was analyzed for structural properties via a 100 ns molecular dynamics (MD) study. Due to the structural shifts and the removal of an open channel potentiator molecule ivermectin from the template structure helical packing changes were observed in the transmembrane segment. Namely removal of ivermectin molecule caused a closure around the Leu 9 position along the ion channel. In terms of the structural shifts there are three potential disulfide bridges between the M1 and M3 helices of the gamma(2) and 2 alpha(1) subunits in the model. The effect of these disulfide bridges was investigated via monitoring the differences in root mean square fluctuations (RMSF) of individual amino acids and principal component analysis of the MD trajectory of the two homology models-one with the disulfide bridge and one with protonated Cys residues. In all subunit types RMSF of the transmembrane domain helices are reduced in the presence of disulfide bridges. Additionally loop A loop F and loop C fluctuations were affected in the extracellular domain. In cross-correlation analysis of the trajectory the two model structures displayed different coupling in between the M2-M3 linker region protruding from the membrane and the beta 1-beta 2/D loop and cys-loop regions in the extracellular domain. Correlations of the C loop which collapses directly over the bound ligand molecule were also affected by differences in the packing of transmembrane helices. Finally more localized correlations were observed in the transmembrane helices when disulfide bridges were present in the model. The differences observed in this study suggest that dynamic coupling at the interface of extracellular and ion channel domains differs from the coupling introduced by disulfide bridges in the transmembrane region. In STZ-induced diabetes atrophy of the thymus is caused by elevated intracellular calcium levels leading to apoptosis. Hyperglycemia is known to result in a decrease in numbers of T cells in the thymus and circulation. Intracellular calcium levels increase in diabetic animals after induction by STZ. Hyperglycemia inhibits Ca2+-ATPase and increases intracellular calcium levels. We have investigated apoptosis in thymus tissue of neonatal STZ (n-STZ)-diabetic rats and the effects of isradipine as a calcium channel blocker (CCB) on apoptosis. Five groups of newborn Wistar rats were used. On the second day after birth 100mg/kg STZ was given i.p. to the first two groups. The first group was n-STZ diabetic. To the second group starting from the 12th week 5 mg/kg/day isradipine (i.p) was given for 6 weeks. To the third group the same dose of isradipine was given on the second day followed by STZ treatment. The fourth group was non-diabetic and treated with 5 mg/kg/day isradipine for six weeks. The fifth group consisted of non-diabetic rats. To the sixth group dexamethasone (5 mg/kg i.p.) was given to adult rats. For detection of apoptotic cells in paraffin-embedded thymus sections the terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick end Labelling (TUNEL) assay was used. The DNA ladder method was performed for anaLysis of DNA fragmentation. In the isradipine-treated non-diabetic group typical apoptotic banding patterns were found whereas thick bands between 123 and 246 bp length were found in the n-STZ- and n-STZ+isradipine-treated groups. More apoptotic cells were observed in the thymus of isradipine-treated n-STZ-treated and n-STZ+isradipine-treated groups when compared with the non-diabetic control and isradipine+n-STZ-treated groups. In conclusion we observed that long-term STZ diabetes results in apoptosis in the thymus. Microtubule dynamics are characterized by dynamic instability-the ability of individual microtubules to switch between phases of growth and shrinkage. Dynamic instability can be explained by the GTP-cap model, suggesting that a "cap" of GTP-tubulin subunits at the growing microtubule end has a stabilizing effect, protecting against microtubule catastrophe-the switch from growth to shrinkage. Although the GTP-cap is thought to protect the growing microtubule end, whether the GTP-cap size affects microtubule stability in cells is not known. Notably, microtubule end-binding proteins, EBs, recognize the nucleotide state of tubulin and display comet-like localization at growing microtubule ends, which can be used as a proxy for the GTP-cap. Here, we employ high spatiotemporal resolution imaging to compare the relationship between EB comet size and microtubule dynamics in interphase LLC-PK1 cells to that measured in vitro. Our data reveal that the GTP-cap size in cells scales with the microtubule growth rate in the same way as in vitro. However, we find that microtubule ends in cells can withstand transition to catastrophe even after the EB comet is lost. Ece; Goksun, TilbePreterm birth increases the likelihood of early language and cognitive delays, but less is known about later aspects of language development, such as narrative generation. Narrative skills involve dimensions, such as linguistic and narrative complexity, and preterm (PT) and full-term (FT) children's narrative performances may vary across these dimensions. We investigated the role of neonatal status on the total number of words produced, linguistic complexity, and narrative complexity across two presentation modes: narrative generation while seeing pictures and narrative generation after watching an animated video. Seventy-one Turkish-reared preschool-aged children (31 PT [Mage = 48.70, SD = 1.53] and 40 FT [Mage = 48.83, SD = 1.63]) participated in the study. Despite having lower expressive vocabulary skills (assessed by a standardized task) than full-term children, preterm children performed comparably in both picture and animated video-stories, except PT children tended to produce longer narratives in the picture story, possibly due to the different demand characteristics of the tasks. It has been historically significant in traditional medicine. In our research, we delved into the therapeutic attributes of its oil, emphasizing its antioxidant, antimicrobial, and antitumor properties. The primary chemical constituent of the gum is styrene, accounting for 78.5 %. The gum demonstrated antioxidant capabilities in several assays, including in 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2?-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), cupric reducing antioxidant capacity (CUPRAC) and ferric reducing antioxidant power (FRAP). It displayed bactericidal actions against various gram-positive bacteria, such as Staphylococcus aureus, and gram-negative strains, including Escherichia coli. Additionally, the oil showcased potent antitumor effects against breast (MDA-MB-231), lung (A549), and prostate (PC3) cancer cell lines. These effects were found to be both time- and dose-dependent. L. orientalis Mill. oil showed the best antitumor activity against breast, lung, and prostate cancer cell lines after the 24 h and 48 h treatment. Its oil might induce autophagy in the PC3 prostate cancer cell line, whereas its cytotoxicity against MDA-MB-231 and A549 cancer cell lines might not be correlated with autophagy or apoptosis pathways. In this study, it is aimed to make predictions with machine learning in order to determine the optimal treatment option for prostate cancer patients. The study included 88 male patients diagnosed with prostate cancer. Independent variables were determined as Gleason scores, biopsy, PSA, SUVmax, and age. Prostate cancer treatments, which are dependent variables, were determined as hormone therapy(n = 30), radiotherapy(n = 28) and radiotherapy + hormone therapy(n = 30). Machine learning was carried out in the Python with SVM, RF, DT, ETC and XGBoost. Metrics such as accuracy, ROC curve, and AUC were used to evaluate the performance of multi-class predictions. The model with the highest number of successful predictions was the XGBoost. False negative rates for hormone therapy, radiotherapy, and radiotherapy + hormone therapy treatments were, respectively, 12.5, 33.3, and 0%. The accuracy values were computed as 0.61, 0.83, 0.83, 0.72 and 0.89 for SVM, RF, DT, ETC and XGBoost, respectively. The three features that had the greatest influence on the treatment model prediction for prostate cancer with XGBoost were biopsy, Gleason score (3 + 3), and PSA level, respectively. According to the AUC, ROC and accuracy, it was determined that the XGBoost was the model that made the best estimation of prostate cancer treatment. As a result of overexcitation of NMDARs, neuronal death occurs and may lead to diseases such as epilepsy, stroke, Alzheimer's disease, and Parkinson's disease. In this study, human GluN1- GluN2A type NMDAR structure is modeled based on the X-ray structure of the Xenopus laevis template and missing loops are added by ab-initio loop modeling. The final structure is chosen according to two different model assessment scores. To be able to observe the structural changes upon ligand binding, glycine and glutamate molecules are docked into the corresponding binding sites of the receptor. Subsequently, molecular dynamics simulations of 1.3 mu s are performed for both apo and ligand-bound structures. Structural parameters, which have been considered to show functionally important changes in previous NMDAR studies, are monitored as conformational rulers to understand the dynamics of the conformational changes. Moreover, principal component analysis (PCA) is performed for the equilibrated part of the simulations. From these analyses, the differences in between apo and ligand-bound simulations can be summarized as the following: The girdle right at the beginning of the pore loop, which connects M2 and M3 helices of the ion channel, partially opens. Ligands act like an adhesive for the ligand-binding domain (LBD) by keeping the bi-lobed structure together and consequently this is reflected to the overall dynamics of the protein as an increased correlation of the LBD with especially the amino-terminal domain (ATD) of the protein.Article Citation - Scopus: 13Mechanical strength of antibiotic-impregnated bone cement on Day 0 and Day 15: a biomechanical study with Surgical Simplex P and teicoplanin(2002) Gögüş, A.; Akman, S.; Göksan, S.B.; Bozdag, E.OBJECTIVES: To determine the maximum amount of a specific antibiotic added to 40 g acrylic bone cement in terms of the effect on mechanical properties of the bone cement on Day 0 and after antibiotic release on Day 15. METHODS: In an experimental design, eight teicoplanin doses (0, 400 mg, 800 mg, 1200 mg, 1600 mg, 2000 mg, 3200 mg and 4000 mg) were added to bone cement (Surgical Simplex P). Specimens were prepared using the third generation cementing technique to determine the compressive strength and four-point bending strength according to ASTM and ISO standards, respectively. For each concentration, 10 samples were prepared for Day 0 and Day 15. Mechanical tests were performed on Day 0 and after antibiotic leaching in water at 37 degrees C on Day 15. 0 mg specimens served as controls and mechanical strengths for each antibiotic concentration on Day 0 and Day 15 were compared. RESULTS: In compression tests, Day 0 samples showed no significant differences, whereas Day 15 samples starting with 800 mg exhibited significant decreases in compressive strength. However, the compressive strengths were above the minimum standard of 70 MPa set by ASTM at all concentrations and in all groups. Four-point bending tests demonstrated significant decreases in strength starting with 1200 mg in Day 0 samples, and with 400 mg in Day 15 samples. Four-point bending strengths of 2000 mg, 3200 mg, and 4000 mg samples decreased below, or approximated closely the minimum standard of 50 MPa set by ISO on Day 15. CONCLUSION: Our results suggest that the maximum amount of teicoplanin dose to be safely added to 40 g of Surgical Simplex P is 1600 mg when third generation cement mixing and application techniques are employed.Article Citation - WoS: 5The surgical treatment of atrial fibrillation(Turkish Soc Cardiology, 2007) Akpınar, Belhhan; Sağbaş, Ertan; Güden, Mustafa; Sanisoğlu, İlhanThe surgical treatment of atrial fibrillation (AF) has entered a new era with the development of new tools and advanced techniques. The Maze III operation remains as the gold standard for the surgical treatment of AF. However new energy sources have been adopted for treating this arrhythmia in an effort to reduce the invasiveness and technical concerns with the original procedure. Success rates ranging between 70-98% have been reported using these new techniques. On the other hand interventional cardiologists have further improved their techniques so that percutaneous techniques are competing with minimally invasive ablation techniques for the treatment of drug resistant and symptomatic lone AF. These developments have aroused the interest of cardiac surgeons in AF surgery and have found themselves a wide application. It has been estimated that thirty thousand patients have undergone surgical ablation for AF so far. Inevitably procedure related complications have occurred and caused skepticism by some groups. However surgical ablation has become a widely accepted treatment modality for AF patients undergoing concomitant cardiac surgery. The concept of beliefs could provide a basis for how donors may perceive recipients' end-stage liver failure (ESLF) and surgery for organ donation. However there is no such quantitative study. Therefore the objective of this study was to explore beliefs of living donors about recipients' ESLF and surgery for organ donation. Methods. The sample comprised 16 living donors who donated a part of their liver to a patient who had ESLF. The data were analyzed by following established procedures for inductive qualitative analysis. Results. Analysis showed that donors' beliefs can be viewed in a number of groups. Beliefs about recipients' ESLF included diverse explanations for ESLF (blaming oneself and physicians) and physical symptoms (developmental slowing down). Beliefs about being a donor included reasons for being a donor (performing a good deed being healed) barriers to being a donor (other people being ignorant and selfish) ways to manage these barriers (following one's gut feeling) and factors facilitating being a donor (the feeling that one does not have many people to leave behind). Beliefs about surgery for organ donation included physical effects (pain feeling stiff). Beliefs about organ donation included views that general organ donation should be encouraged and that people's awareness should be raised. Conclusions. Existing psychological perspectives could help to interpret some beliefs. Nevertheless other beliefs not previously reported could be considered as targets for individual consultations/psycho-educational programs for fostering emotional well-being.Article Multi-Omics Profiling Uncovers LINC00486-Associated LncRNA Regulation in Human Traumatic Brain Injury(Genetics Society of Korea, 2025) Al-Rubaye, T.; Isa, Z.; Erenkol, D.; Tarahomi, E.; Erdogan, N.S.Background : Traumatic brain injury (TBI) induces broad molecular changes in the human brain, altering gene expression in diverse neural and glial cells. While the transcriptional effects of TBI on protein-coding genes are well characterized, the roles of long noncoding RNAs (lncRNAs), key regulators of gene expression and chromatin, remain largely unknown. Objective : Our objective was to identify lncRNAs altered in TBI and explore their potential regulatory functions. Methods : We applied an integrative multi-omics approach combining single-nucleus RNA sequencing (snRNA-seq), isoform-level transcriptomics, transposable element (TE) annotation, and RNA-binding protein (RBP) interaction analyses. Public snRNA-seq datasets from cortical tissues of 12 TBI patients and 5 controls were analyzed to resolve injury-driven transcriptional signatures. We have performed differential expression analysis on 12,801 human lncRNAs, examined isoform-specific expression with TE content, and explored RBP–lncRNA interactions using CLIP-seq data. Results : Cell-type diversity decreased in TBI, and reactive and progenitor-like states were expanded. We identified 190 upregulated lncRNAs, mainly in glial cells. Among these, LINC00486 emerged as a brain-enriched lncRNA consistently increased after TBI. Isoform analysis showed its dominant brain isoform contains LINEs and LTRs, linking it to regulatory networks associated with endogenous retroelement activation. Functional enrichment connected LINC00486 to neurodevelopment, serotonin metabolism, and neuroinflammatory pathways. CLIP-seq data confirmed its interactions with stress-responsive RBPs such as AGO2 and TARDBP. Conclusions : Our multi-omics analysis identifies LINC00486 as a potential regulator of transcriptional plasticity in TBI. Oxidative stress mechanisms are often reported to be implied in type 2 diabetes mellitus. In order to determine their clinical relevance we investigated several plasma indicators in the Turkish patients with NIDDM: (1) homocysteine (Hey) and cysteine (Cys) which contribute to increase the risk of atherosclerosis during NIDDM (ii) glutathione (GSH) and cysteinylglycine (CysGly) resulting from GSH degradation catalyzed by gamma-glutamylcysteine transferase (GGT) (iii) malonaldehyde (MDA) as a marker for lipid peroxidation and (iv) total antioxidant status (TAS). Our main results were evaluated based on sex and diabetic status. In female patients plasma concentrations of MDA and Hey were significantly higher than in controls while GSH levels were significantly lower. In males a difference between control and diabetic groups was noticed only for Hey levels being also higher in patients. In the diabetic group increase in serum glucose concentration was significantly correlated with increased GGT activity. In both controls and diabetic patients GGT activity was correlated with a raised Cys concentration and a decreased GSH level. In both controls and diabetic patients there were significant positive correlations between Cys and Hey and between GSH and Hey. We concluded that GSH and MDA levels are clinical indicators for an oxidative process linked to type 2 diabetes mellitus especially in women.Article Citation - WoS: 1Impact of Culture Shock, Acculturation Approaches, and Sociocultural Adaptation on Perceived Proficiency in Turkish and English Among International Students in Turkey(Elsevier, 2025) Tekel, Esra; Ergin-Kocaturk, Hatice; Su, Ahmet; Kocaturk, Metin; Caner, Mustafa; Karadag, EnginThe primary objective of this study was to investigate the influence of culture shock, acculturation strategies, and sociocultural adjustment on the perceived Turkish and English language proficiency of international students in Turkey. Data were collected from 3554 international students in Turkey whose native language was neither Turkish nor English, using the "Culture Shock Questionnaire", "Acculturation Measurement", and "Sociocultural Adjustment Scale". Data were analyzed using correlation and hierarchical multiple regression analyses. The results revealed that the perceived Turkish and English language proficiencies of international students in Turkey were moderate. Language proficiency, personal interests, and social participation factors of sociocultural adjustment and acculturation strategies, such as assimilation and integration, have a positive impact on perceived Turkish language proficiency. On the other hand, the interpersonal stress factor of culture shock and assimilation and integration aspects of the acculturation strategies were found to be the most critical determinants of perceived English language proficiency. Additionally, language proficiency, personal interests, and social participation factors of sociocultural adjustment and acculturation strategies, such as separation and assimilation, had positive effects, whereas the language proficiency factor of sociocultural adjustment and assimilation acculturation strategies had negative effects. The findings of this study indicate that acculturation strategies and sociocultural adjustment play a crucial role in the development of perceived target language proficiency.
