Abstract

In the present study, we have investigated the distribution of ErbB receptors and the characteristics of cell proliferation in breast cancer cell lines MDA MB 231 and T47D by immunocytochemical analysis. In MDA MB 231 cells, ErbB1 (EGFR) and ErbB2 receptors were stained in the nucleus, while in T47D cells they were detected in the cell membrane. The distribution of ErbB3 receptors was similar but less dense than the ErbB2 receptors in both cell lines. The distribution of tyrosine phosphorylation was also found similar to ErbB3receptors. In addition, when we investigated the level of cellular proliferation with PCNAexpression and BrdU incorporation, we found that the rate of cell proliferation in MDA MB231 cells was 2.5 fold higher than T47D cells.In conclusion, the nuclear localization of ErbB family receptors in MDA MB 231 and their higher rate of proliferation indicate the malignant phenotype of these cells compared toT47D cells. In addition, in the presence of ErbB receptor expression, the low level of tyrosine phosphorylation suggests the involvement of other factors in the regulation of cell proliferation in these cells.
