This new method uses a probabilistic approach that itself uses Bayesian learning to estimate the network coefficients. A posterior probability density function (PDF) is composed using the likelihood and prior PDFs derived from the system model and prior information respectively. This posterior PDF is used to draw samples with the help of the Metropolis algorithm a special case of the Metropolis--Hastings algorithm where the proposal distribution function is symmetric and resulting samples are then averaged to find the minimum mean square error (MMSE) estimate of the network coefficients. Active and inactive conformations revealed by recent crystallographic experiments do not provide a complete dynamic picture of the receptor especially in the binding site. In this study molecular dynamics (MD) simulation through a residue-based coarse-grained model is used as an alternative and efficient method to explore a wider conformational search space. The system was composed of beta(2)AR embedded into a 1-palmitoyl-2-oleoyl-phosphatidylcholine membrane bilayer with surrounding water. A total of 6 mu s of simulation at constant NPT was performed for a system of 6868 coarse-grained beads. The system reached equilibrium at around 0.1 mu s. The overall 3-dimensional structure was well preserved throughout the simulation. Local residue-based fluctuations were in good agreement with fully atomistic MD simulations. Four distinct snapshots were selected and reverse-mapped to all-atom representations with around 65000 atoms. Each reverse-mapped system was later subjected to 100 ns of MD simulation for equilibration. Root mean square deviation clustering analysis yielded distinct receptor conformers for the binding site regions which were suggested to be alternative representations of the binding pocket and thus were proposed as plausible targets in docking-based virtual screening experiments for the discovery of novel antagonists.Article İki Dillilik Deneyimi ile İki Dilli Çocukların Bilişsel Kontrolü Arasındaki İlişki(Dilbilim Derneği, 2024) Gong, Xıaoweı; Aktan-ercıyes, Aslı; Karadöller, Dilay Z.İkidilli kişilerin çeşitli bilişsel yetileri ile İkidillilik deneyiminin farklı boyutları arasında nasıl ilişkili olduğuna dair giderek artan bir ilgi bulunmaktadır. Ancak, bu ilginin yeterli dil gelişimi olmayan küçük yaştaki ikidilli çocuklardan veri elde etme konusunda sınırlı kaldığı görülmüştür. Mevcut çalışma, 30-48 aylık ikidilli çocukların ve ebeveynlerinin dil yeterliği, dil kullanımı, ve kod değiştirme sıklığı gibi çeşitli ikidillilik deneyimine dair faktörleri incelemektedir. Sonuçlar, çocukların bilişsel kontrol yeteneklerini, çocuklardan veya ebeveynlerden gelen ikidilli deneyim değişkenlerinden ziyade, çocukların yaşının yordadığını göstermiştir. Bu çalışma, üçlü ikidillilik deneyimi boyutlarını ve ebeveyn faktörlerini de içererek ikidilliğin etkileri inceleyen az sayıdaki çalışmadan biridir. Its active site shows a resemblance to those of 2 homologous enzymes monamine oxidase A and B (MAO-A and MAO-B.) In the present work starting from suitable scaffolds and generating thousands of structures from them 10 potential inhibitors were obtained with structural and physicochemical properties selectively suitable for inhibiting LSD1. iLib Diverse software was used to generate the diverse structures and 3 docking tools CDOCKER GOLD and AutoDock were used to find the most probable potential inhibitor based on its binding affinity. The dispositions of the candidate molecules within the organism were checked by ADMET_PSA_2D (polar surface area) versus ADMET_AlogP98 (the logarithm of the partition coefficient between n-octanol and water) and their suitability is discussed. The LSD1 inhibition activities of the candidates were compared with the properties of trans-2-phenylcyclopropylamine (tranylcypromine) and 2-(4-methoxy-phenyl) cyclopropylamine which are the 2 known inhibitors of LSD1.Article The Influence of the Belgian Constitution on Ottoman Constitutional Law;(Istanbul University Press, 2024) Akad,M.The Belgian Constitution of 1831 is one of the constitutions that set an example during the preparation process of Ottoman constitutional law. The most important reason for this was the spirit of the Belgian Constitution, which brought together the monarch, parliament, and executive branch. Despite being inspired by the Belgian Constitution, Ottoman constitutional law was significantly different. The main objective of this study is to identify the similarities and differences between the two documents and to understand the spirit of Ottoman constitutional law. Therefore, to clarify the differences between documents, a comparative method is adopted primarily centered on articles that organize the structure and authority of the monarch and legislative and executive bodies. The main differences can be observed in the relationship among these three bodies. When the necessary provisions are compared, Ottoman constitutional law lacks specific provisions that might allow the development of equitable harmony between the monarch, parliament, and cabinet. More precisely, Ottoman constitutional law did not have the foundations for limiting the sultan’s power and creating a balanced division of power among the three bodies. Consequently, despite their similarities, Ottoman constitutional law lacks certain elements that allow it to be called a constitution; therefore, it is instead referred to as a constitutional charter. © 2024 Istanbul University Press. Thus, a novel compound, SH2U was synthesized and its ability for the inhibition of human MAO (hMAO) activity was investigated by our group. In addition, the interaction of SH2U with hMAO isoforms have been investigated in detail using molecular modelling technics. It has been found that SH2U inhibited hMAO-B potently, selectively, competitively and reversibly suggesting that the novel compound may be a promising drug agent for the treatment of Parkinson's and Alzheimer's diseases. Methods: 1-(3,5-dichloro-2-hydroxyphenyl)-3- p-tolylprop-2-ene-1-on (3',5'-Dichloro-2'-hydroxy- 4-methyl chalcone) was prepared via the reaction of p-tolualdehyde and 3',5'-Dichloro-2'-hydroxy acetophenone in methanol in the presence of KOH. Then, the obtained chalcone was treated with isonicotinic acid hydrazide under reflux in ethanol to give (3-(3,5-Dichloro-2-hydroxy phenyl)-5-p-tolyl- 4,5-dihydropyrazol-1-yl) (pyridin-4-yl) methanone. The interaction of SH2U with hMAO isoforms was investigated fluorometrically using commercial kits. The interaction between SH2U and hMAO was also analyzed using AutoDock 4.2.6 program. Results: The structure of compound SH2U was confirmed using IR, Mass, 1 H-NMR and elemental analysis methods. SH2U appeared as a potent, selective, reversible and nontoxic hMAO-B inhibitor. Mode of inhibition was found to be competitive. Interactions of the new compound with the active site of hMAO-B were clarified using molecular modelling studies. Conclusion: Compound SH2U inhibited hMAO-B potently, selectively, competitively and reversibly. The synthesized compound is found to be more potent and selective than selegiline, the known irreversible MAO-B inhibitor, indicating that SH2U appears as a promising active molecule to be used in the treatment of Parkinson's and Alzheimer's diseases. © 2018 Refik Saydam National Public Health Agency (RSNPHA).Amaç: Nöromediatörlerin yikimindan sorumlu olan monoamin oksidaz (MAO) enziminin izoformlarinin (MAO-A ve -B) birçok hastalik ile yakindan ilişkili olduğu; MAO inhibitörlerinin depresyon, Parkinson ve Alzheimer hastaliği gibi hastaliklarin tedavisinde kullanildigi bilinmektedir. Grubumuzca daha etkin, tersinir ve az yan etkili yeni bir MAO inhibitörü (SH2U bileşiği) sentezlenmiş ve bu bileşiğin insan MAO enzimini (hMAO) inhibe etme yeteneği incelenmiştir. Ayrica bu yeni bileşiğin hMAO ile etkileşimi, moleküler modelleme çalişmalari ile detayli bir şekilde araştirilmiştir. Sentezlenen yeni bileşiğin hMAO'yu kuvvetli bir şekilde yarişmali ve tersinir olarak inhibe ettiği bulunmuştur. Söz konusu bileşiğin Parkinson ve Alzheimer hastaliklarinin tedavisinde ümit verici bir ilaç etken maddesi olabileceği düşünülmektedir. Yöntem: 3',5'-Dikloro-2'-hidroksi asetofenon ile p-tolualdehit'in metanol içinde KOH varliginda reaksiyona girmesiyle 1-(3,5-dikloro-2-hidroksifenil)- 3-p-tolil prop-2-en-1-on (3',5'-Dikloro-2'-hidroksi-4- metil şalkon) bileşiği sentez edilmiştir. Daha sonra elde edilen bu bileşiğin etanol içerisinde geri çeviren sogutucu altinda izonikotinik asit hidrazit ile muamele edilmesiyle [3-(3,5-dikloro-2-hidroksifenil)-5-p-tolil-4,5- dihidropirazol-1-il] (piridin-4-il) metanon bileşiği sentez edilmiştir. Yapisi doğrulanan bu bileşiğin hMAO enzimi ile etkileşimi, ticari tayin kiti kullanilarak fluorometrik bir yöntemle incelenmiştir. Ayrica, söz konusu yeni bileşik ile hMAO arasindaki etkileşimler, moleküler modelleme çalişmalari ile aydinlatilmiştir. Bulgular: Sentezlenen bileşiğin yapisi, IR, Mass, 1H-NMR ve elemental analiz yöntemleri kullanilarak doğrulanmiştir. Yapisi doğrulanan bu bilesigin etkin, seçici, tersinir, toksik olmayan bir hMAO-B inhibitörü olduğu ve inhibisyonun yarişmali olduğu görülmüştür. Moleküler yerleştirme programi kullanilarak bileşiğin hMAO-B enziminin aktif bölgesinde hangi amino asit yan zincirleri ile ne tür girişimleri yaptiği belirlenmiştir. Sonuç: Yeni sentezlenen SH2U bileşiği, hMAO-B enzimini kuvvetle, seçici, yarişmali ve tersinir olarak inhibe etmistir. The decision version of the problem, List k-Coloring, is NP-complete even for bipartite graphs. As an application of list coloring problem we are interested in the Futoshiki Problem. Futoshiki is an NP-complete Latin Square Completion Type Puzzle. Considering Futoshiki puzzle as a constraint satisfaction problem, we first give a list coloring based algorithm for it which is efficient for small boards of fixed size. To thoroughly investigate the efficiency of our algorithm in comparison with a proposed backtracking-based algorithm, we conducted a substantial number of computational experiments at different difficulty levels, considering varying numbers of inequality constraints and given values. All synthesized compounds were characterized by FTIR,1H-NMR,13C-NMR and MS data. In vitro cytotoxic activities of all synthesized molecules against five cancer lines (human breast cancer MCF-7, human lung cancer A549, human prostate cancer PC-3, human chronic myelogenous leukemia K562, human neuroblastoma SH-SY5Y cell lines) were evaluated by MTT assay. Compounds were also tested on mouse embryonic fibroblast cells (NIH/3T3) to determine selectivity. Eighteen target compounds 9-26 were screened for their mPGES-1 and COX-1/2 inhibitory activities. Of these compounds, 26 (KUC16D425) showed the highest mPGES-1 inhibition at 10 mu M. This compound has also been observed to induce apoptosis and inhibit cell migration in MCF-7 cells. In silico molecular docking calculations were performed to understand the binding interactions of compounds with target proteins. People with diabetes who smoke have a significantly higher risk of morbidity and mortality. Smoking greatly increases diabetes complications. Surveys were conducted to ascertain how frequently nurses counseled patients to quit and their interest in cessation training. Observations of nurse- patient interactions and interviews with diabetes nurses facilitated the development of a culturally sensi- tive cessation training program. Survey results revealed that diabetes nurse educators recognized the need for training in smoking cessation, as many patients with diabetes do not consider smoking to be harm- ful for their health. A two-day workshop was developed for nurses on the specific harms of smoking for diabetes patients, including hands-on training in cessation counseling and motivational interviewing. Two months after training, nurses reported that the skills they acquired gave them confidence to counsel patients but identified four challenges to conducting cessation in their clinical settings. Diabetes nurse educators can play a pivotal role in delivering cessation. Training and overcoming challenges to implementation are urgently needed in the time of coronavirus disease 2019 given the dual risks of diabetes and smoking for severe complications.
